• Preparation: This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, human cell-derived, recombinant human PD1 extracellular domain (rhPD1; Catalog#10377-H08H; aa 1 - 167; NP_005009.2). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

• Specificity: Human PD1 / PDCD1 / CD279
• No cross-reactivity with human cell lysate ( 293 cell line ) in WB and ELISA
• Western blot: This antibody can be used at 1-2 μg/mL with the appropriate secondary reagents to detect human PD1 in WB. Using a DAB detection system, the detection limit for human PD1 is approximately 8 ng/lane under non-reducing conditions and 4 ng/lane under reduced conditions.
• Direct ELISA: This antibody can be used at 0.5-1μg/mL with the appropriate secondary reagents to detect human PD1. The detection limit for human PD1 is approximately 1.25 ng/well
• Storage: This antibody can be stored at 2℃-8℃ for 1 month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20℃ to -70℃. Preservative-Free.
  Sodium azide is recommended to avoid contamination (final concentration 0.05%-0.1%). It is toxic to cells and should be disposed of properly. Avoid repeated freeze-thaw cycles.

PD1 / PDCD1 / CD279 Antibody Background

Programmed Cell Death 1 (PD1) is a typeⅠtransmembrane immunoreceptorer and belongs to the CD28 immunoglobulin superfamily that mediates immune responses. PD1 is expressed on activated T cells, B cells, myeloid cells and a subset of thymocytes. PD1 inhibits the T cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN-γ by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. PD1 has been suggested to be involved in lymphocyte clonal selection and peripheral tolerance, and thus contributes to the prevention of autoimmune diseases. Furthermore, PD1 is shown to be a regulator of virus-specific CD8+ T cell survival in HIV infection.
