Quantitative determination of Human 2B4 / CD244
1. Capture Antibody 0.5 mg/mL of rabbit anti-2B4 monoclonal antibody (in PBS, pH 7.4). Dilute to a working concentration of 2 μg/mL in CBS before coating. (Catalog: # 10042-R025)
2. Detection Antibody 0.25 mg/mL rabbit anti-2B4 monoclonal antibody conjugated to horseradish-peroxidase (HRP) (in PBS, 50 % HRP-Protector, pH 7.4). Dilute to working concentration of 0.0625 μg/mL in detection antibody dilution buffer before use. (Catalog: # 10042-R031)
3. Standard Each vial contains 56 ng of recombinant 2B4. Reconstitute with 1 mL detection antibody dilution buffer. After reconstitution, store at -20℃ to -80℃ in a manual defrost freezer. A seven-point standard curve using 2-fold serial dilutions in sample dilution buffer, and a high standard of 8 ng/mL is recommended.

2B4 / CD244 Background Information
The CD244 antigen, also known as 2B4, is a cell surface glycoprotein implicated in the regulation of natural killer and T lymphocyte function. 2B4 is a member of the signaling lymphocyte activation molecule (SLAM)-related receptor family and is important for stimulating NK cell cytotoxicity and cytokine production, which is expressed on all NK cells, a subpopulation of T cells, monocytes and basophils. The 2B4 antigen identified on NK cells and T cells is capable of transmitting stimulatory signals and non-MHC-restricted killing. Reported as an activating receptor, human 2B4 can effectively activate and enhance NK cell–mediated cytotoxicity, induce secretion of IFN-γ and matrix metalloproteinases (MMPs), as well as NK cell invasiveness.
