Immunofluorescence staining of NCAPH in U2OS cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human NCAPH polyclonal antibody (dilution ratio 1:500) at 4℃ overnight. Then cells were stained with the Alexa Fluor®594-conjugated Goat Anti-rabbit IgG secondary antibody (red). Positive staining was localized to Nucleus.

Non-SMC condensin I complex subunit H (NCAPH) in colon cancerous tissues was higher than that in all corresponding adjacent non-cancerous tissues.NCAPH is highly expressed in colorectal cancer cell lines comparing with normal human colonic epithelial cells, and identified many NCAPH mutations in colon cancer (CC) patients.Depletion of NCAPH inhibits CC cell proliferation, migration in vitro and xenograft tumor formation in vivo.NCAPH knockdown promotes cell apoptosis and cell cycle arrest at G2/M phase.NCAPH could be used as a new therapeutic target in CC in future.
