Immunofluorescence staining of EGFR in monkey skin with rabbit monoclonal antibody (1:50, frozen section). The image showing membrane staining of squamous epithelium cell.
Immunochemical staining of human EGFR in human uterine cervix cancer with rabbit monoclonal antibody (1:200, formalin-fixed paraffin embedded sections). Positive staining was localized to uterine gland.
Immunochemical staining of human EGFR in human rectal cancer with rabbit monoclonal antibody (1:200, formalin-fixed paraffin embedded sections). Positive staining was localized to rectal gland. The left panel: tissue incubated with primary antibody; The right panel: tissue incubated with the mixture of primary antibody and antigen (recombinant protein).
Immunochemical staining of human EGFR in human placenta with rabbit monoclonal antibody (1:200,formalin-fixed paraffin embedded sections). Positive staining was localized to trophoblast.
Immunofluorescence staining of EGFR in A431 cells. Cells were fixed with 4% PFA,blocked with 10% serum, and incubated with rabbit anti-human EGFR monoclonal antibody (dilution ratio 1:60) at 4℃ overnight. Then cells were stained with the Alexa Fluor®488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Cytoplasm and Cell membrane.
As a member of the epidermal growth factor receptor (EGFR) family, EGFR protein is type I transmembrane glycoprotein that binds a subset of EGF family ligands including EGF, amphiregulin, TGF-α, betacellulin, etc. EGFR protein plays a crucial role in signaling pathway in the regulation of cell proliferation, survival and differentiation. Binding of a ligand induces EGFR protein homo- or heterodimerization, the subsequent tyrosine autophosphorylation and initiates various down stream pathways (MAPK, PI3K/PKB and STAT). In addition, EGFR signaling also has been shown to exert action on carcinogenesis and disease progression, and thus EGFR protein is proposed as a target for cancer therapy currently.
